Kapal pembinasa Kelas Arleigh Burke merupakan sebuah kapal pembinasa peluru berpandu, dan salah satu daripada kelas-kelas kapal pembinasa yang terdapat di dalam Tentera Laut Amerika Syarikat. Ia merupakan kapal pembinasa pertama yang dibina berdasarkan sistem tempur Aegis dan radar jujukan fasa pelbagai fungsi SPY-1D. Kapal pertama dari kelas ini telah ditauliahkan pada 4 Julai 1991. Selepas pelucutan tauliah kapal terakhir dari Kelas Spruance, iaitu USS Cushing, pada 21 September, 2005, kapal pembinasa Kelas Arleigh Burke menjadi satu-satunya kelas kapal pembinasa yang masih aktif di dalam Tentera Laut A.S.
Kelas ini dinamakan sempenan Laksamana Arleigh "31-Knot" Burke, seorang pegawai kapal pembinasa Amerika Syarikat yang paling terkenal semasa Perang Dunia II. Laksamana Burke masih hidup apabila kapal pertama dari kelas ini ditauliahkan.
